Utilization of Electrophoresis in Testing the Genetic Purity of F1 Hybrid Seeds of Brassica Campestris L. ssp . pekinensis


Abstract
This experiment was conducted to develop a method for testing the degree of purity of the F©û hybrid variety in Chinese cabbage by separating patterns of phosphoglucomutase (PGM) and phosphoglucose isomerase (PGI) isozymes extracted from the imbibed seeds.
1. Starch gel electrophoresis techniques were used to separate isozymes of PGM and PGI of Chinese cabbage.
2. Diagnostic isozyme band variants of PGM-2 were found in the two parents of the hybrid A, and those of PGI-1 were in the parents of the hybrid B.
3. F©ü and backcross progeny analysis data revealed that PGM-2 and PGI-1 variants are controlled by co-domiant alleles, PGM-2 F/F and PGM-2 S/S, PGI-1 S/S and PGI-1 F/F, repectively.
